柯尊記,男,1996年于中山大學(xué)醫(yī)學(xué)院解剖學(xué)與神經(jīng)科學(xué)專業(yè)獲博士。先后在中國科學(xué)院上海腦研究所,美國伊利偌斯大學(xué)醫(yī)學(xué)院解剖學(xué)與細(xì)胞生物學(xué)系和美國康奈爾大學(xué)醫(yī)學(xué)院神經(jīng)科學(xué)系從事博士后研究。1998-1999年任湖北醫(yī)藥學(xué)院副教授,解剖學(xué)教研室主任。2003年底加入中國科學(xué)院中國科學(xué)院上海生命科學(xué)研究院營養(yǎng)科學(xué)研究所,2004年入選中國科學(xué)院“百人計(jì)劃”。2006年入選上海市“浦江人才計(jì)劃”?,F(xiàn)任中國科學(xué)院上海生命科學(xué)研究院營養(yǎng)科學(xué)研究所研究員,博士研究生導(dǎo)師,神經(jīng)營養(yǎng)科學(xué)研究組組長。

研究內(nèi)容

長期從事衰老的神經(jīng)科學(xué)機(jī)制研究,發(fā)現(xiàn)了氧化代謝紊亂、氧自由基產(chǎn)生增加、免疫/炎癥反應(yīng)是維生素B1缺乏(TD)引起的選擇性中線旁丘腦核神經(jīng)元死亡的重要病理生理過程;內(nèi)質(zhì)網(wǎng)應(yīng)激、未折疊和錯(cuò)誤折疊蛋白累積、caspase-12激活是參與代謝性氧化損傷引起的神經(jīng)元死亡的主要細(xì)胞機(jī)制;證實(shí)神經(jīng)元產(chǎn)生的單核細(xì)胞趨化因子,是氧化代謝紊亂誘發(fā)免疫/炎癥反應(yīng)的開關(guān)分子。發(fā)現(xiàn)氧化代謝紊亂可以通過增強(qiáng)β分泌酶的活性增加β淀粉樣蛋白的產(chǎn)生,從而加速神經(jīng)元病變的發(fā)展過程。TD可增加淀粉樣蛋白的產(chǎn)生,亞臨床維生素B1缺乏是阿爾茲海默癥的危險(xiǎn)因素之一。

研究領(lǐng)域

1)中西醫(yī)結(jié)合防治神經(jīng)退行性疾病。在傳統(tǒng)中藥中尋找可以延緩老年性神經(jīng)退行性疾病發(fā)展的生物活性小分子和中藥復(fù)方。為防治老年性神經(jīng)退行性疾病,尤其是阿爾茨海默病提供策略和方法。

2)闡明老年性神經(jīng)元死亡的可能細(xì)胞、分子生物學(xué)機(jī)制,尋找能客觀反映老齡健康的生物學(xué)標(biāo)記,和延緩老年性神經(jīng)元死亡的分子靶點(diǎn)。

3)探討酒精濫用等環(huán)境因素對神經(jīng)系統(tǒng)發(fā)育和衰老影響的細(xì)胞、分子生物學(xué)機(jī)制。
